Abstract
Objective: To examine the effect of different concentrations of sucrose (50 or 100 mM) and omega-3 (0.5 or 1%) on post- thawed quality of ram semen cryopreserved in tris extender. Methods: Semen samples were diluted in tris extender, tris + sucrose (50 and 100 mM/L) to ensure 60 million motile spermatozoa mL-1, cooled slowly up to 5 C and equilibrated for 2 h. Semen was packed into 0.25 mL cryovial. The cryovials were placed horizontally on a rack and frozen in a vapor 4 cm above liquid nitrogen (LN2) for 15 minutes then dipped in liquid LN2. Frozen cryovial were thawed media (containing different concentrations of omega-3) at 37C for 1 min. The parameters studied were sperm motility, sperm membrane integrity (WT), DNA fragmentation and mitochondrial apoptosis. Results: The output data demonstrated that addition of 50–100 mM of sucrose/L after thawing had significantly (P<0.01) ameliorated motility, membrane integrity, DNA fragmentation and mitochondrial apoptosis % compared to control diluted semen, while the binary interaction between sucrose and omega-3 concentration on the post-thawing sperm parameters there is a high significant effect (p> 0.01) of interaction on sperm motility, it was found that the highest value was belonged to (T3\omega-31%) (38.71%) in comparison with control. Membrane integrity (WT) was affected with interaction the highest value was from (T3\omega-3, 1%) (43.14%) in comparison with control. Also, mitochondrial apoptosis trait was affected with the same binary interaction there the highest value was shown from control (T1\ omega-3 0%) (83.85%), while the lowest one was belong sucrose (T2\omega-3 0.5%) (74%). Finally, binary interaction have effect on DNA fragmentation trait, shows the highest value which was come from control (T1\ omega-30%) (7.07) treatment in comparison with lowest one (T2\omega-3 0.5%) (5.50%).
